在此基礎上，根據國內外已發表的ibv基因序列，分別設計特異性引物，應用不同引物進行反轉錄合成cdna ，分片段對ibv的主要結構基因進行pcr擴增，並分別將各個目的片段克隆到puc19載體上，在大腸桿菌dh5中實現目的基因的分子克隆，經藍白斑篩選、限制性內切酶分析、 pcr鑒定，篩選出重組陽性質粒，並對各個目的基因片段進行序列測定，從而獲得ibv主要結構基因全序列。
