首先，構建了ecbp21表達載體，誘導了重組蛋白的表達，並通過膠回收法獲得了大量純化重組ecbp21蛋白，制備了高效價、高特異性抗體；隨后，利用ecbp21抗體，結合免疫膠體金電鏡定位技術進行了ecbp21亞細胞定位研究，結果顯示：在白芷愈傷組織細胞和花序軸細胞中金顆粒主要分佈在細胞壁區域，而在細胞內未發現或僅有少量金顆粒分佈，表明ecbp21蛋白主要定位於細胞壁區域，這為細胞外cambp （ ecbp21 ）的胞外存在提供了直接證據：進一步，利用ecbp21抗體，通過免疫組織化學分析研究了ecbp21組織特異性分佈狀況，結果表明ecbp21在白芷各組織中均有分佈，但在葉、花、花序軸中分佈較多，而在葉柄、根中分佈較少。
