本實驗從人胎盤中提取總rna ，利用rt - pcr技術獲得了編碼jo - 1的基因整長序列，選用impact - cn系統中的ptyb11載體，構建了jo - 1基因的克隆與表達載體，並轉化大腸桿菌er2566 ，經過抗性篩選、分子量大小比較、雙酶切鑒定、和pcr鑒定等多種方法驗證，篩選出了5個陽性克隆。
