繼代培養時，經過反復實驗比較雞胚原代成纖維細胞飼養層（ pcef ） 、小鼠原代成纖維細胞飼養層（ pmef ） 、 snl細胞飼養層等三種不同飼養層的作用效果，最終發現以雞胚原代成纖維細胞製作飼養層同時添加各種生長因子最適合雞類eg細胞的生長，但pcef與pmef之間的差異不顯著。
