本實驗採用pcr方法從番茄花cdna文庫中克隆到葉綠體shsp基因，經測序證實與genbank中已發表的序列在編碼區相差2個堿基，其中一個堿基導致1個氨基酸的改變。將葉綠體shsp基因定向克隆于帶有組成性表達啟動子camv35s的植物表達載體prok中，凍融法轉化農桿菌lba4404 ，利用葉圓盤法對番茄進行ti質粒介導的遺傳轉化。
