從4 ? 8周的流產胎兒的原始生殖嵴部位分離到原始生殖細胞，經過機械和化學方法的解離后培養於事先用絲裂黴素處理過的小鼠胚胎成纖維細胞或人胚胎成纖維細胞飼養層上，培養基中添加了lif ， bfgef ， foskolin等細胞因子，此後大約每7天左右傳代一次。
